ABSTRACT

The scalp is the structure that covers the skull. It is commonly affected by painful processes resulting from infestations, infectious or inflammatory diseases. This pain located in the scalp does not have well-defined clinical characteristics and is not yet included in the ICHD-3 diagnostic criteria. The authors suggest including this pain in the next classification of headaches as a headache attributed to a scalp disorder.

ABSTRACT

Abstract The objective of this paper was to develop and evaluate two semi-solid pharmaceutical forms containing 0.1% tacrolimus: cream (CRT01) and gel (GLT01). For the evaluation of physicochemical stability, at times 0, 30, 60 and 90 days, at 23°C and at 40°C, High Performance Liquid Chromatography coupled with a Diode Array Detector (HPLC-DAD) was employed. This method was developed and validated for tacrolimus quantification. The occlusivity test and skin permeation assay were also performed, using an animal model (Wistar rats), and the CRT01 and GLT01 were compared to the 0.1% tacrolimus ointment (PFU01) obtained from the University Pharmacy, Federal University of Rio de Janeiro, Brazil. CRT01 and GLT01 presented a homogeneous aspect and consistency adequate for topical products, along with sensory characteristics above PFU01. They also presented adequate physicochemical stability for 90 days and a lower occlusive effect than PFU01 (p<0.05). CRT01 showed greater affinity for the skin when compared to PFU01 and GLT01, with low systemic absorption. The CRT01 semi-solid formulation was considered the most adequate one to treat patients with atopic dermatitis or other dermatologic inflammatory diseases, promoting rational use of tacrolimus

ABSTRACT

Abstract Syphilis is a disease with compulsory and mandatory notification to the Notifiable Diseases Information System (SINAN), with benzathine benzylpenicillin being the treatment of choice. The aim of the study was to compare the consumption of benzylpenicillin benzathine, from the dispensation, between the health regions of a capital in the southern region of the country, according to the georeferencing of notified cases of syphilis. This is a descriptive, cross-sectional, retrospective study of the use of benzylpenicillin benzathine and of reported cases of syphilis. Data on syphilis cases were obtained from notifications made in SINAN, and drug consumption data were obtained from the Municipal Health Department computerized system for Drug Dispensing from January 1st, 2019 to December 31st, 2019. Notifications and drug consumption were georeferenced according to 8 health regions. From the compilation of data, the rates of cases and consumption in relation to the population of each region were calculated. A total of 3188 notifications and a total of 35191 vials of benzathine benzylpenicillin were analyzed. The ratio of vials by SINAN notifications showed that each patient took 11 vials of the drug, which is a higher value if we consider that the complete treatment is 2 to 6 vials per case.

ABSTRACT

Abstract Periodontitis is an oral disease associated with inflammation and pain with swollen and bleeding gums. In the present study, dental pastes containing NSAIDs, namely, diclofenac sodium and nimesulide (1 % w/w) were prepared to treat periodontitis. Dental pastes of diclofenac sodium and nimesulide (1 % w/w) were prepared with/without mucoadhesive hydrocolloid polymers such as sodium carboxy methyl cellulose (NaCMC), hydroxyl ethyl cellulose (HEC) and methyl cellulose (MC) by conventional trituration method. The pH, drug content, viscosity, tube spreadability and tube extrudability of these prepared dental pastes were measured. These dental pastes of diclofenac sodium and nimesulide (1 % w/w) were characterized by FTIR analyses for drug-excipient compatibility. The in vitro drug releases from these dental pastes in 6.4 pH phosphate buffer solution displayed sustained release over longer period and the drug release rate was found to be decreased when the concentration of mucoadhesive polymer was increased. These dental pastes displayed good adhesion to the oral mucosa revealing more retention time in mouth when tested for ex vivo mucoadhesion using bovine cheek pouch. The stability study results reveal that the DC3 and NC3 dental paste formulations were found stable enough over a longer period in different storage conditions. The present study revealed that the prepared mucoadhesive dental pastes of diclofenac sodium and nimesulide (1 % w/w) had good adhesion with the oral mucosa to maintain consistent release of drugs over prolonged time.

ABSTRACT

The use of plants in disease treatment is cost effective and relatively safe. This study was designed to investigate anti-hyperlipidemic and anti-diabetic activity of ethanolic leaf extract of Catharanthus roseus alone and in combination therapy in hyperlipidemic & diabetic mice. Eight groups comprising five mice each were used. Group A was hyperlipidemic control, group B, C, D received atorvastatin (20 mg/kg), leaf extract (200 mg/kg) and leaf extract in combination with atorvastatin (200 mg/kg and 20 mg/kg) orally for 15 days. Group E was diabetic control. Group F, G, H received sitagliptin (40 mg/kg), leaf extract (200 mg/kg) and extract in combination with sitagliptin (200 mg/kg and 40 mg/kg) orally for 7 days. Blood cholesterol levels were measured at 1st, 5th, 10th and 15th day and fasting blood sugar levels were measured at 2, 12, 24, 72 and 168 hours during treatment. One-way ANOVA with tukey- kramer multiple comparison test was used. The chemical characterization of ethanolic extract of Catharanthus roseus leaves showed presence of alkaloids, saponins, tannins and flavonoids. Ethanolic extract of Catharanthus roseus has significant anti-hyperlipidemic & anti-diabetic effects (p<0.05, p<0.01) when compared with control but had not cause significantly increase in anti-hyperlipidemic effects of atorvastatin. While significantly increased the antidiabetic effect of sitagliptin (p<0.05)

ABSTRACT

This article aimed at describing the history of insertion of the clinical condition currently referred to as "obesity" in the eleven revisions of the International Classification of Diseases (ICD) published by the World Health Organization (WHO) between 1900 and 2018. For this purpose, a search for the keyword obesity was performed in the documents, with subsequent description and analysis of the presence, mode of insertion, and changes occurring over time. The results demonstrated that the condition has been and continues to be inserted as symptom, morbidity, comorbidity, cause of mortality and/or disease. It can be concluded that there is a massive logical inconsistency in the principles that govern the classification.

ABSTRACT

ABSTRACT

ABSTRACT

ABSTRACT The disease related to IgG4 (ER-IgG4 in Spanish) is a multi-systemic, autoimmune disease characterized by inflammatory fibro-sclerotizing lesions forming masses or pseudo tumors, high concentrations of serum IgG4 and extensive tissue infiltrations by IgG4-positive plasma cells. It includes several entities previously considered organ-specific ones. It can affect almost any organ, being pancreas, salivary glands, lacrimal glands, lymphatic ganglion, retroperineum and kidneys the most affected ones. The diagnosis is made through combining clinical, serologic, imaging and histopathological findings. It has a reserved prognosis but with an early diagnosis its control is reached, and occasionally also the disease’s remission, although it tends to have relapses. It is an underdiagnosed disease, partially due to its recent characterization. We present two cases entering the range covered by this disease, the Riedel’s thyroiditis and retroperitoneal fibrosis, both initially answering to therapy with high doses of corticoids after diagnosis (AU).

ABSTRACT

The atopic dermatitis (DA by its acronym in Spanish) is a chronic inflammatory disease and its etiopathogenesis is multifactorial. The diversity of risk factors described in the bibliography evidences the complicatedness of the research with these aims and the interrelation among them offers a problem for the precise approach in each case, given the overlapped effects found. The aim is to give an up-dated review on atopic dermatitis etiopathogenesis and physiopathology helping to increase and up-date information on this topic. A review was carried out in articles appeared mainly in PubMed and Scielo databases during the last five years, and the results were systematized to contribute to the professional growth of health professionals working in the primary health care (AU).

ABSTRACT

The pharmaceutical industry is increasingly joining chemoinformatics in the search for the development of new drugs to be used in the treatment of diseases. These computational studies have the advantage of being less expensive and optimize the study time, and thus the interest in this area is increasing. Among the techniques used is the development of multitarget directed ligands (MTDLs), which has become an ascending technique, mainly due to the improvement in the quality of treatment involving several drugs. Multitarget therapy is more effective than traditional drug therapy that emphasizes maximum selectivity for a single target. In this review a multitarget drug survey was carried out as a promising strategy in several important diseases: neglected diseases, neurodegenerative diseases, AIDS, and cancer. In addition, we discuss Computer-Aided Drug Design (CADD) techniques as a tool in the projection of multitarget compounds against these diseases.

ABSTRACT

Adhesion is a widely conserved mechanism of virulence among Gram-negative bacteria that is used by several pathogens to colonize their hosts, being the first step in biolfilm development. A variety of appendages and proteins are involved in bacterial adhesion, such as pili, fimbriae, fimbrial and afimbrials adhesins. The phytopathogen Xylella fastidiosa, causal agent of important diseases such as Pierce's disease of grapevines, citrus variegated chlorosis and olive quick decline syndrome, harbours on its surface several of these structures that are potentially responsible for efficient colonization of insect vectors and plant hosts. Among the afimbrial adhesins encoded in the genome of this bacterium, three XadAs (XadA1, Hsf/XadA2 and XadA3) are classified as trimeric autotransporters. Data from the literature suggest that XadA1 and XadA2 are important for biofilm formation, but XadA3 function has not been yet investigated. In this work, we aimed to biochemically and functionally characterize the XadA3 protein and gather additional information about the role played by XadA1 and XadA2 in X. fastidiosa adhesion and virulence. Using immunodetection with a polyclonal anti-XadA3 antibody, we have demonstrated that this protein localizes to the bacterial surface and mediates intercellular adhesion. Phenotypic characterization of the deletion mutants of XadA adhesins encoded genes revealed that the ΔxadA3 mutant has reduced cell aggregation capacity and biofilm formation when compared to both ΔxadA1 and ΔxadA2 mutants as well as to Temecula wild type strain. Deletion of the xadA genes marginally affects the global gene expression profile assessed by RNA-seq of the mutant strains compared to the wild-type strain, eventhough an increase in lipase/esterase transcripts levels was observed in the mutant strains. It has been reported that these enzymes appear to participate in the degradation of plant tissue that is associated with symptoms of Pierce's disease of grapevines. The deletion of xadA3 results in a phenotype of hypervirulence in grapevines but also of deficiency in insect-vector transmission. The results obtained in this work evidenced the important role played by XadAs adhesins, particularly XadA3, in X. fastidiosa intercellular adhesion, biofilm development and virulence

ABSTRACT

Physical exercise improves physical fitness and older adults? health conditions, there is doubts whether this can compensate the harmful effects of that participate in health promotion programs with osteoarticular diseases. The objective was to evaluate the physical fitness of older adults participating in community groups considering the practice of regular exercise and the presence of osteoarticular diseases. It is a cross-sectional study in which 891 elderly participants in exercise programs and educational/cultural activities of public institutions were recruited. They were divided into four groups according to physical exercise and the presence of osteoarticular diseases. A research tool identified sociodemographic characteristics, and presence of osteoarticular diseases (osteoarthritis, rheumatoid arthritis and osteoporosis). Anthropometric measurements (weight, height, waist and hip) and physical fitness (test battery - American Alliance for Health, Physical Education, Recreation and Dance) were applied. Kruskal Wallis, Mann Whitney and chi-square tests were used, p <0.05. Older adults who presented osteoarticular diseases and practiced exercise had better physical fitness than those with similar diseases but not exercise practitioners and the older adults without osteoarticular diseases and practiced exercise had better physical fitness than those without diseases but not exercise practitioners. The coordination, agility/dynamic balance and strength resistance showed significant differences among the groups, where the group who practiced physical exercise had better scores. This study indicates that physical exercise can reduce the functional deficits resulting from osteoarticular diseases of elderly that participate in health promotion programs.

ABSTRACT

Tumor incidence in children is lower than in adults. In fetuses and newborns it is even rarer. We report a series of three cases of germ cell tumors; the first one present in anewborn child, the second one in a stillborn, and a third tumor present in placenta membranes. We also present the results of a review of the literature of similar cases of tumors...
